It is much faster than it sounds. All you need to do is
(1) In Photoshop, cut a rectangular region containing the hotspot
feature. Notice the top left coordinate.
(2) Select the hotspot feature, eg using the magic wand or
other selection tools.
(3) Fill the selection with white color, and save as gif.
If you just need the rectangle, omit step (2) and (3),
and fill the whole rectangle with white color.

I think this masking of hotspots can be used in very powerful
ways, eg to mark complicated features, or people etc.
